SmartAb™ Recombinant Anti-JAG1 pH-dependent Antibody (Clone J1J22)

The anti-JAG1 pH-dependent antibody (clone J1J22) targets human JAG1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-JAG1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced JAG1 clearance. The antibody operates in a cyclical manner, binding JAG1 in the extracellular fluid and releasing it in acidified endosomes. Human JAG1 (Jagged 1) is a transmembrane protein that acts as a ligand for the Notch receptor. It is a key player in the Notch signaling pathway, which is critical for cell fate determination, proliferation, and differentiation, and is implicated in various developmental disorders and cancers.
