SmartAb™ Recombinant Anti-LGALS9 pH-dependent Antibody (Clone TIB-141)

The anti-LGALS9 pH-dependent antibody (clone TIB-141) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human LGALS9. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for LGALS9 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from LGALS9 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human LGALS9 (Galectin-9) is a lectin that acts as an immune checkpoint molecule by binding to TIM-3 on T-cells, which can lead to T-cell exhaustion and apoptosis. It is a key regulator of immune responses and a potential target for cancer immunotherapy.
