SmartAb™ Recombinant Anti-LTA pH-dependent Antibody (Clone ch2C8)

The anti-LTA pH-dependent antibody (clone ch2C8) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human LTA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for LTA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from LTA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human LTA (Lymphotoxin-alpha) is a cytokine of the TNF family that is essential for the development of lymphoid organs and plays a role in inflammatory responses. Its dysregulation is implicated in autoimmune diseases, making it a therapeutic target.
