The anti-PLAU pH-dependent antibody (VS-0825-YC285) targets human PLAU in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-PLAU acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced PLAU clearance. The antibody operates in a cyclical manner, binding PLAU in the extracellular fluid and releasing it in acidified endosomes. Human PLAU (Plasminogen Activator, Urokinase), also known as uPA, is a serine protease that is involved in the degradation of the extracellular matrix. It plays a critical role in cell migration and tissue remodeling, and its overexpression is strongly associated with cancer metastasis.
