SmartAb™ Recombinant Anti-SLPI pH-dependent Antibody (Clone 43H7)

The anti-SLPI pH-dependent antibody (clone 43H7) targets human SLPI in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-SLPI acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced SLPI clearance. The antibody operates in a cyclical manner, binding SLPI in the extracellular fluid and releasing it in acidified endosomes. Human SLPI (Secretory Leukocyte Peptidase Inhibitor) is a protease inhibitor that plays a crucial role in the innate immune response as a potent anti-inflammatory molecule. It inhibits enzymes such as neutrophil elastase and is a potential therapeutic target for inflammatory diseases.
