The anti-TNFRSF14 pH-dependent antibody (clone 44A9) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Tumor Necrosis Factor Receptor Superfamily, Member 14 (TNFRSF14). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF14 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF14 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF14, also known as HVEM, is a receptor for the ligands LIGHT and BTLA. It is involved in T-cell co-stimulation and is a key immune regulator, making it a potential therapeutic target for cancer immunotherapy.
