anti-HIgG(Fc)Fab-C-DMDM ADC

This ADC product is composed of a Fab fragment of an anti-human IgG Fc specific polyclonal antibody conjugated via a cleavable linker to DMDM (anti-human IgG(Fc)Fab-CL-DMDM). The antibody portion is a Fab fragment of a secondary antibody and the drug portion, duocarmycin DM (DMDM), is a cytotoxic small molecule which causes cell death by binding to the minor groove of DNA and alkylating the adenine. This product displays no obvious toxicity without a primary antibody and can be a quite efficient and economical alternative to pre-screening human monoclonal antibodies as ADC candidates.

Economical Screening Tool
This secondary ADC has been a fantastic alternative to producing custom conjugates. We used it to screen our panel of human antibodies. The anti-human IgG Fc Fab fragment bound our primary antibodies well, and the DMDM payload provided a clear cytotoxic readout.

Effective DNA Minor Groove Binder
The Duocarmycin DM payload is a potent DNA alkylating agent. We observed cell death consistent with DNA minor groove binding. This product allowed us to quickly assess the internalization potential of our antibody candidates without extensive synthesis.
