CAMK2D: A Central Mediator of Cardiac Disease

Calcium/Calmodulin-Dependent Protein Kinase II Delta (CAMK2D) is the major CAMKII isoform expressed in the heart. It is an important calcium signal transducer, playing a fundamental role in normal excitation-contraction coupling in the heart. During pathological conditions such as chronic stress (e.g., heart failure, myocardial infarction), CAMK2D becomes pathologically hyperactive. The kinase aberrantly phosphorylates key cardiac calcium-handling proteins (e.g., ryanodine receptor, phospholamban) causing arrhythmias, cardiomyocyte death, and adverse cardiac remodeling. Thus, the development of pharmacologic strategies to inhibit CAMK2D is an important and active area of therapeutic development.
