CCR3: The Eosinophil Chemotactic Receptor

C-C Motif Chemokine Receptor 3 (CCR3) is a G-protein coupled receptor (GPCR) that is most highly expressed on eosinophils, a central player in allergic inflammation. It is also expressed on basophils as well as certain T-cell subsets. CCR3 has high affinity for the chemokines CCL11 (eotaxin-1), CCL24 (eotaxin-2) and CCL26 (eotaxin-3). The binding of these chemokines provides the primary signal to recruit eosinophils from the circulation into the tissue of interest, such as the airways in asthma or the esophagus in eosinophilic esophagitis. Eosinophils and the toxic granule proteins they release are a major cause of pathology in these diseases, which is why CCR3 is a major therapeutic target.
