CDC42: A Master Regulator of Cell Polarity and the Cytoskeleton

CDC42 is a member of the Rho family of small GTPases, which function as molecular switches cycling between an inactive GDP-bound form and an active GTP-bound form. In the active state, it interacts with diverse effector proteins to regulate a host of fundamental cellular processes. It is most well-known for its role as a master regulator of cell polarity, defining the axis of migration and organizing the cytoskeleton. It does this primarily through the activation of effector kinases that regulate the actin-nucleating Arp2/3 complex, resulting in the generation of filopodia and lamellipodia, the protrusions responsible for cell movement. Its activity is required for everything from wound healing to neuronal development.
