Description: Noncleavable linker is considered noncleavable-meaning linker cleavage, and payload release does not depend on the differential properties between the plasma and some cytoplasmic compartments. Instead, the release of the cytotoxic drug is postulated to occur after internalization of the ADC via antigen-mediated endocytosis and delivery to lysosomal compartment, where the antibody is degraded to the level of amino acids through intracellular proteolytic degradation.

Humanized Antibody For STEAP1
The humanized antibody clone 120v.24 showed excellent binding to the six-transmembrane epithelial antigen of the prostate (STEAP1). We are studying its expression at cell-cell junctions, and this reagent has been critical for our localization and inhibition studies.
Consistent MMAF Inhibition
The MMAF payload linked via the noncleavable MC linker provided consistent results in our cytotoxicity assays. The drug was released upon lysosomal degradation of the antibody, leading to the expected accumulation of cells in metaphase arrest due to tubulin inhibition.
