Through immunohistochemistry, FACS analysis, confocal microscopy, and cell transfection studies, the researchers demonstrate that labyrinthin not only serves as a marker but appears functional in the cancer phenotype, with its expression promoting cancerous characteristics in normal cells and its inhibition normalizing cancer cell morphology.
Creative Biolabs provided recombinant human IgG1 (MHH-78) antibodies that bind to ASPH in HEK 293 cells as well as polyclonal antibodies (MOR-0269) targeting ASPH amino acids 250-350. These antibodies helped the researchers distinguish between ASPH and labyrinthin binding patterns, clarifying the cellular localization and expression patterns of these proteins and establishing labyrinthin as a distinct cell-surface target for potential adenocarcinoma immunotherapy approaches.
