The anti-ACVR2B pH-dependent antibody (VS-0825-YC208) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human ACVR2B. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for ACVR2B under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from ACVR2B at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. ACVR2B (Activin Receptor Type-2B) is a receptor for activin and myostatin, which are members of the TGF-β superfamily and are key regulators of muscle growth. Antagonizing ACVR2B is a therapeutic strategy being explored for muscle wasting diseases.
