SmartAb™ Recombinant Anti-C1s pH-dependent Antibody (VS-0825-YC223)

The anti-C1s pH-dependent antibody (VS-0825-YC223) targets both human and Cynomolgus C1s in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-C1s acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced C1s clearance. The antibody operates in a cyclical manner, binding C1s in the extracellular fluid and releasing it in acidified endosomes. Human C1s is a serine protease that is a key component of the classical pathway of the complement system, playing a critical role in innate immunity.
