SmartAb™ Recombinant Anti-CD200R1 pH-dependent Antibody (Clone huDX182)

The anti-CD200R1 pH-dependent antibody (clone huDX182) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD200R. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD200R under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD200R at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD200R on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD200R is a receptor for CD200, primarily found on myeloid cells, that delivers an inhibitory signal upon binding. This interaction is crucial for maintaining immune homeostasis and preventing excessive inflammation.
