Cumulative data from at least six animals per condition (a). Total number of human cells (CD19+) out of 105 events is shown. (b) Homing of freshly purified PBMCs from CLL patients was studied using the same model. Data were analyzed by morphological gating on lymphocytes (R1) and subsequently on the CD45+ subset (R2). The CD19/CD5 fluorescence levels were then determined for cells contained in R3 (R1 × R2). (c) Pre-incubation with blocking anti-CD38 mAb (SUN-4B7) negatively interferes with homing of CLL lymphocytes to spleen and BM. Anti-CXCR4 mAb was used as the positive control, whereas an isotype-matched (IgG1) anti-CD38 mAb (OKT10) was used as a binding but nonblocking control mAb. Results are presented as mean±s.e.m. of eight independent experiments, with each condition analyzed in duplicate.

Competitive binding assays demonstrate blocking of Alexa-647–conjugated OKT10 mAb binding to CD38-expressing L363 multiple myeloma cells with escalating concentrations of unlabeled OKT10-FP (A) or OKT10-CC (B). No blocking is observed with escalating concentrations of nonbinding control Ab constructs.

Radiation absorbed doses to tumor, whole body, and normal tissues are estimated by integrating the areas under time–activity curves constructed by plotting the concentration of 111In–DOTA–biotin (after OKT10-CC pretargeting) measured in tissues by γ counting at 2, 24, 48, and 96 hours (33).
