The anti-CD38 pH-dependent antibody (VS-0825-YC54) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD38. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD38 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD38 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD38 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD38 is a transmembrane glycoprotein expressed on a variety of immune cells, most notably plasma cells. It is a key marker and therapeutic target in multiple myeloma, where it is highly expressed on malignant plasma cells.
