The anti-CD48 pH-dependent antibody (VS-0825-YC62) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD48. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD48 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD48 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD48 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD48 is a cell surface glycoprotein expressed on immune cells, particularly lymphocytes and monocytes. It is a ligand for CD244 (2B4) and plays a role in co-stimulating T-cells and NK cells, thereby contributing to immune activation.
