The anti-CXCL10 pH-dependent antibody (VS-0825-YC90) targets human CXC motif chemokine 10 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-CXCL10 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced CXCL10 clearance. The antibody operates in a cyclical manner, binding CXCL10 in the extracellular fluid and releasing it in acidified endosomes. Human CXCL10, also known as Interferon-gamma-inducible protein 10 (IP-10), is a chemokine that chemoattracts monocytes, T-cells, and NK cells. It is strongly induced by interferon-gamma and is a critical player in the immune response against viruses and in autoimmune diseases.
