The anti-FCGR3A pH-dependent antibody (clone M/YC-27) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human FCGR3A. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for FCGR3A under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from FCGR3A at pH 6.0. This pH-sensitive antibody retains its binding affinity for FCGR3A on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human FCGR3A, also known as CD16a, is a low-affinity receptor for the Fc region of IgG antibodies. It is expressed on NK cells, macrophages, and neutrophils, and its binding to antibody-coated cells is crucial for antibody-dependent cellular cytotoxicity (ADCC).
