The anti-FGFR1 pH-dependent antibody (VS-0825-YC131) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and monkey FGFR1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for FGFR1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from FGFR1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for FGFR1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. FGFR1, or Fibroblast Growth Factor Receptor 1, is a receptor tyrosine kinase that binds to FGF ligands. It is involved in cell proliferation, differentiation, and development, and its dysregulation is linked to various cancers and developmental disorders.
