SmartAb™ Recombinant Anti-GDF15 pH-dependent Antibody (Clone 3B7-3-3)

The anti-GDF15 pH-dependent antibody (clone 3B7-3-3) targets human Bone morphogenetic protein-9 (BMP9) in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-BMP9 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced BMP9 clearance. The antibody operates in a cyclical manner, binding BMP9 in the extracellular fluid and releasing it in acidified endosomes. Human Bone morphogenetic protein-9 (BMP9) is a secreted protein that belongs to the TGF-β superfamily. It is a potent inducer of bone formation and is being investigated as a therapeutic agent for bone regeneration.
