The anti-GPNMB pH-dependent antibody (clone Glembatumumab) is a pH-responsive antibody engineered to target human GPNMB. This pH-switched monoclonal antibody (mAb) was developed by incorporating histidine residues into its variable regions, which results in pH-biased binding to GPNMB. By leveraging the natural pH variations in different tissues and cellular compartments, this pH-responsive antibody facilitates precise targeting of GPNMB on cells and tissues. Human GPNMB, or Glycoprotein NMB, is a transmembrane protein that is involved in cell adhesion and is a target for immunotherapy in melanoma and other cancers. Its expression is associated with a poor prognosis in various tumors.
