The anti-IL5 pH-dependent antibody (VS-0825-YC204) targets human IL5 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL5 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. These histidine substitutions within its Complementarity-Determining Regions (CDRs) enable pH-responsive antigen binding, which leads to enhanced IL5 clearance. The antibody operates in a cyclical manner, binding IL5 in the extracellular fluid and releasing it in acidified endosomes. IL-5 (Interleukin-5) is a cytokine that promotes the growth, differentiation, and activation of eosinophils. It is a key mediator in allergic inflammation and host defense against parasitic infections, playing a critical role in the pathogenesis of asthma and other eosinophil-driven diseases.
