The anti-IL5RA pH-dependent antibody (VS-0825-YC205) targets human IL5RA in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL5RA acid-switched antibody was engineered by incorporating histidine residues into its variable regions. These histidine substitutions within its Complementarity-Determining Regions (CDRs) enable pH-responsive antigen binding, which leads to enhanced IL5RA clearance. The antibody operates in a cyclical manner, binding IL5RA in the extracellular fluid and releasing it in acidified endosomes. IL5RA (Interleukin-5 Receptor Subunit Alpha) is a component of the IL-5 receptor complex, which mediates the signaling of the cytokine IL-5. It is essential for the function of eosinophils and is a therapeutic target for eosinophil-driven inflammatory diseases.
