The anti-LOXL2 pH-dependent antibody (VS-0825-YC240) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human LOXL2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for LOXL2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from LOXL2 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human LOXL2 (Lysyl Oxidase Like 2) is an extracellular enzyme that cross-links collagen and elastin. It is involved in tissue fibrosis, wound healing, and is often overexpressed in cancer, where it promotes tumor invasion and metastasis.
