The Anti-PDGFRA Antibody Prodrug, Protease Activated (2.175.3) is an antibody prodrug comprised of a recombinant human antibody targeting human and mouse PDGFRA, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to PDGFRA protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to PDGFRA effectively. PDGFRA (Platelet-Derived Growth Factor Receptor Alpha) is a receptor tyrosine kinase that becomes an oncogenic driver through specific gene fusions or activating mutations (e.g., D842V). The most notable example is its role in gastrointestinal stromal tumors (GIST), where it drives cell proliferation. It is effectively treated with targeted TKIs.

- This gene encodes a cell surface tyrosine kinase receptor for members of the platelet-derived growth factor family. These growth factors are mitogens for cells of mesenchymal origin. The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. Studies suggest that this gene plays a role in organ development, wound healing, and tumor progression. Mutations in this gene have been associated with idiopathic hypereosinophilic syndrome, somatic and familial gastrointestinal stromal tumors, and a variety of other cancers.
