SmartAb™ Recombinant Anti-PDGFRA pH-dependent Antibody (Clone Olaratumab)

The anti-PDGFRA pH-dependent antibody (clone Olaratumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human PDGF-R alpha (PDGFRA). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for PDGFRA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from PDGFRA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human PDGF-Rα (PDGFRA) is a receptor tyrosine kinase that binds to PDGF ligands. Its signaling pathway regulates cell growth, proliferation, and migration, and its dysregulation is linked to various cancers and fibrotic diseases.
