The anti-PDGFRB pH-dependent antibody (VS-0825-YC284) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human PDGFRB. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for PDGFRB under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from PDGFRB at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human PDGFRB (Platelet-Derived Growth Factor Receptor Beta) is a receptor tyrosine kinase that binds to PDGF ligands. It is a key mediator of cell growth, proliferation, and angiogenesis, and its dysregulation is implicated in cancer development.
