The anti-SELP pH-dependent antibody (clone Inclacumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Selectin P. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SELP under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SELP at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human Selectin P (SELP), also known as CD62P, is a cell adhesion molecule expressed on activated endothelial cells and platelets. It is a key mediator of leukocyte rolling and recruitment to inflammatory sites.
