The anti-VIM pH-dependent antibody (clone 3F9-1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Vimentin. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for Vimentin under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from Vimentin at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human Vimentin is an intermediate filament protein and a key marker for mesenchymal cells. It plays a central role in cell migration and is a hallmark of the epithelial-to-mesenchymal transition (EMT), a process associated with cancer metastasis.
