ACTB: A Ubiquitous Cytoskeletal Regulator in Cancer Neurodegeneration and Developmental Disorders

Beta-actin, encoded by the ACTB gene, is a fundamental and highly conserved component of the cytoskeleton, playing a vital role in preserving cell shape, enabling movement, and facilitating intracellular transport. Alterations in its expression or function have been increasingly linked to a broad range of diseases. In cancer, its dysregulation promotes tumor growth by enhancing cell migration, invasion, and modifying adhesion properties, thereby supporting metastasis. Genetic variants influencing ACTB expression have been found in developmental disorders, especially those involving craniofacial defects and intellectual disabilities, highlighting the importance of cytoskeletal integrity during early development. Immune system disorders may also arise from abnormal beta-actin dynamics, which can hinder immune cell mobility and antigen processing, weakening the body’s defense. Additionally, emerging research connects beta-actin instability to neurodegenerative diseases such as Alzheimer’s and amyotrophic lateral sclerosis, where cytoskeletal breakdown is a hallmark. The protein also plays a key role in tissue repair and wound healing, emphasizing its significance in maintaining normal physiological functions. Background

This gene encodes one of the six actin isoforms, a group of evolutionarily conserved proteins essential for cellular movement, structural support, mechanical integrity, and signaling between cells. The protein product is a key element of the contractile system and belongs to the two non-muscle actins that are broadly expressed across various cell types. Genetic mutations in this gene are linked to Baraitser-Winter syndrome 1, a condition marked by intellectual disability and distinctive craniofacial features. Additionally, multiple pseudogenes related to this gene have been found throughout the human genome.

Component of the BAF complex, which includes at least actin (ACTB), ARID1A, ARID1B/BAF250, SMARCA2, SMARCA4/BRG1, ACTL6A/BAF53, ACTL6B/BAF53B, SMARCE1/BAF57 SMARCC1/BAF155, SMARCC2/BAF170, SMARCB1/SNF5/INI1, and one or more of SMARCD1/BAF60A, SMARCD2/BAF60B, or SMARCD3/BAF60C (PubMed:18765789). In muscle cells, the BAF complex also contains DPF3 (PubMed:18765789). Found in a complex with XPO6, Ran, ACTB and PFN1 (PubMed:14592989). Interacts with XPO6 and EMD (PubMed:15328537). Mouse Anti-ACTB Recombinant Antibody (clone 1D12E12B5) (CAT#: ZG-0296U)
We performed Western blot analysis on human epithelial cell lysates using the Mouse Anti-ACTB Recombinant Antibody. The antibody yielded distinct and precise bands at the anticipated molecular weight, with minimal background noise. It integrated seamlessly with our usual protocol without needing additional adjustments. The signal remained stable across several experiments, facilitating accurate quantification. Overall, it proved to be a dependable choice for detecting beta-actin as a loading control in our protein assays.

Rabbit Anti-ACTB Recombinant Antibody (clone Q20-K), FITC (CAT#: ZG-0423U)
This rabbit anti-ACTB recombinant antibody worked well in my FC with Jurkat and PBMCs, showing clear staining without excessive background. Tested WB on HeLa and HEK293 lysates—sharp bands at ~42kDa. Also tried IHC-P on human colon tissue, giving consistent cytoplasmic staining. Minimal optimization needed for dilutions. Solid performance across cell and tissue samples for ACTB detection.

Recombinant Mouse Anti-ACTB Antibody (clone 8H5-1H4-9E8) (CAT#: VS3-FY31)
Tested this anti-ACTB antibody on FFPE tonsil and colon tissue for IHC-P. Gave crisp cytoplasmic staining at 1:150 dilution with 20min HIER (pH6). Worked consistently across batches, though epithelial cells showed slightly stronger signal than stromal areas. Background was clean with proper blocking. Didn't try other applications, but for routine IHC-P it's been dependable.
