CANX: A Guardian of Glycoprotein Folding in the ER
Calnexin (CANX) is a Type I integral membrane protein of the endoplasmic reticulum (ER) and a member of the molecular chaperones. Calnexin is an important regulator of the N-linked glycoprotein quality control process in the endoplasmic reticulum. It is responsible for recognizing, binding, and retaining monoglucosylated glycans present on newly synthesized polypeptide chains and halting the folding process to prevent aggregation. This cycle is collectively called the "calnexin/calreticulin cycle", where calnexin acts as a central hub for protein quality control alongside its soluble homolog calreticulin and other ER-resident enzymes.

Background
The protein encoded by this gene is a member of the calnexin family of molecular chaperones. The protein is a calcium-binding, endoplasmic reticulum (ER)-associated protein that transiently associates with newly synthesized N-linked glycoproteins, facilitating protein folding and assembly. It may also serve a central role in the quality control of protein folding by sequestering misfolded protein subunits within the ER for degradation. Alternatively spliced transcript variants encoding the same protein have been described.
