CYCS: The Central Switch for Apoptosis

Cytochrome C (CYCS) is a small, highly conserved protein. It has two cellular compartments where it functions physiologically and pathologically. Physiologically, CYCS is a mobile electron carrier found in the intermembrane space of the mitochondria that transports electrons from Complex III to Complex IV (Cytochrome c oxidase). Pathologically, it is the primary initiator of the intrinsic apoptosis pathway. In response to pro-apoptotic stimuli, CYCS is released from the mitochondria into the cytosol where it binds Apaf-1, an essential step in the formation of the apoptosome and the activation of Caspase-9. The cell is now committed to die.
