HSPA8: Potential target in Aging, Alzheimer's Disease, and Lysosomal Storage Disorders
HSPA8, or Hsc70, is a member of the Hsp70 family expressed at high levels under normal conditions. It is the central player in CMA, where it recognizes proteins with a specific motif and delivers them to the lysosome for degradation. CMA activity naturally declines with age, contributing to toxic aggregates in neurodegenerative diseases. Furthermore, HSPA8 is involved in clathrin-mediated endocytosis and the uncoating of coated vesicles. Understanding the regulation of HSPA8 activity is therefore necessary for developing therapies that restore cellular clearance pathways. Robust antibody tools are required to detect HSPA8 in various cellular compartments.

Background
This gene encodes a member of the heat shock protein 70 family, which contains both heat-inducible and constitutively expressed members. This protein belongs to the latter group, which are also referred to as heat-shock cognate proteins. It functions as a chaperone, and binds to nascent polypeptides to facilitate correct folding. It also functions as an ATPase in the disassembly of clathrin-coated vesicles during transport of membrane components through the cell. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
