HSPA8: Potential target in Aging, Alzheimer's Disease, and Lysosomal Storage Disorders

HSPA8, or Hsc70, is a member of the Hsp70 family expressed at high levels under normal conditions. It is the central player in CMA, where it recognizes proteins with a specific motif and delivers them to the lysosome for degradation. CMA activity naturally declines with age, contributing to toxic aggregates in neurodegenerative diseases. Furthermore, HSPA8 is involved in clathrin-mediated endocytosis and the uncoating of coated vesicles. Understanding the regulation of HSPA8 activity is therefore necessary for developing therapies that restore cellular clearance pathways. Background

This gene encodes a member of the heat shock protein 70 family, which contains both heat-inducible and constitutively expressed members. This protein belongs to the latter group, which are also referred to as heat-shock cognate proteins. It functions as a chaperone, and binds to nascent polypeptides to facilitate correct folding. It also functions as an ATPase in the disassembly of clathrin-coated vesicles during transport of membrane components through the cell. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
