IL7R: A Key Transmembrane Protein in Immune System Regulation
Interleukin-7 receptor (IL7R) is a key transmembrane protein involved in immune system regulation. It is primarily expressed on T cells, B cell precursors, and other immune cells, playing a crucial role in lymphocyte development, survival, and homeostasis. IL7R consists of two subunits: IL7Rα (CD127) and the common gamma chain (γc), which together mediate signaling upon binding to its ligand, interleukin-7 (IL-7). This interaction activates the JAK-STAT and PI3K-AKT pathways, promoting cell proliferation, differentiation, and survival. IL7R is essential for proper immune function, and its dysregulation is associated with various diseases, including severe combined immunodeficiency (SCID), autoimmune disorders, and certain cancers such as leukemia. Due to its critical role in immune cell regulation, IL7R has become an important target for therapeutic research, with ongoing studies exploring its potential in immunotherapy and autoimmune disease treatment.

Background
The protein encoded by this gene is a receptor for interleukin 7 (IL7). The function of this receptor requires the interleukin 2 receptor, gamma chain (IL2RG), which is a common gamma chain shared by the receptors of various cytokines, including interleukins 2, 4, 7, 9, and 15. This protein has been shown to play a critical role in V(D)J recombination during lymphocyte development. Defects in this gene may be associated with severe combined immunodeficiency (SCID). Alternatively spliced transcript variants have been found.
