- Peptide linkers, belonging to Enzymatically cleavable linkers, combine greater systemic stability with rapid enzymatic release of the drug in the target cell. The scission of peptidic bonds relies on lysosomal proteolytic enzymes, which have very low activities in blood due to endogenous inhibitors and the unfavorably high pH value of blood.

- Auristatins are water-soluble dolastatin analogs of dolastatin 10. Dolastatin 10 belongs to dolastatin family and it can powerfully bind to tubulin, thus inhibiting polymerization mediated through the binding to the vinca alkaloid binding domain, and causes cell to accumulate in metaphase arrest.

- This gene encodes a pentaspan transmembrane glycoprotein. The protein localizes to membrane protrusions and is often expressed on adult stem cells, where it is thought to function in maintaining stem cell properties by suppressing differentiation. Mutations in this gene have been shown to result in retinitis pigmentosa and Stargardt disease. Expression of this gene is also associated with several types of cancer. This gene is expressed from at least five alternative promoters that are expressed in a tissue-dependent manner. Targeting CD133 On Stem Cells
We used this product to target the stem cell marker CD133 (Prominin-1). The antibody clone AC133 is the standard for this target. The vc linker facilitated the delivery of the MMAF payload, which is a potent microtubule inhibitor.
Robust Enzymatic Drug Release
The valine-citrulline linker cleaved efficiently in our assays involving lysosomal proteolytic enzymes. We observed the accumulation of cells in metaphase arrest, confirming the release and activity of the auristatin F payload in colorectal cancer cells.
