The binding of the antisera (1:100) and MAb (10 mg/ml) to the individual peptides and peptide complexes was detected by dot blot assay. The complex N-36/C-34 was prepared by mixing peptides N-36 and C-34 in PBS at equimo- lar concentrations (2 mM).

Each of the N51(L6)C43, N36(L6)C34, and N34(L6)C28 polypeptides (∼1 μM) were used to coat wells of a microplate. The binding of the purified NC-1 IgG at the indicated concentrations to each coated polypeptide was tested by ELISA.

HIV-1IIIB-infected cells were reacted with the MAbs NC-1 (A) and 2F5 (B) (5 μg of IgG/ml) in the presence or absence of sCD4 (10 μg/ml) and analyzed by flow cytometry. Binding of NC-1 to HIV-2ROD-infected cells was also tested (C). Normal mouse (NM) IgG was used as a control.
