Recombinant human antibody to Human NOTCH2. It is a novel anti-cancer stem cell antibody identified by screening against the Notch3 receptor that binds to and prevents signaling through both the Notch2 and Notch3 receptors. It has been shown in preclinical models to have broad-spectrum anti-tumor activity via inhibition of cancer stem cell growth, and promoting cell differentiation, as well as disrupting tumor angiogenesis by inhibiting vascular pericytes.

Flow cytometry analysis of U87 MG cells with purified antibody TAB-H67 (QCAb249, Blue). Cells were blocked with Human TruStain FcXTM and washed with 0.5% BSA-PBS. Cells were then incubated with TAB-H67 (5 μl/5x10^5 cells) for 60 min at 4°C. The secondary antibody was FITC anti-human IgG antibody (3 μl) for 60 min at 4°C.

SDS-PAGE analysis of TAB-H67 in reduced (Lane 1) and non-reduced (Lane 2) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result, different β-mercaptoethanol-reduced proteins (Heavy chain and Light chain) migrate as about 50 kDa and 25 kDa, respectively.
The purity of TAB-H67 was greater than 95% as determined by SDS-PAGE.

Western blot analysis of TAB-H67 was performed by loading Recombinant NOTCH2 protein.
TAB-H67 incubation concentration: 2 ng/μL.
The secondary antibody: HRP-goat anti-human IgG
Lane 1: Reducing Antigen (0.1 μg)
Lane 2: Reducing Antigen (0.3 μg)
Lane 3: Reducing Antigen (0.6 μg)

ELISA analysis of TAB-H67 was performed by coating with Recombinant Human NOTCH2 Protein. Then blocked with BSA and incubated with Anti-Human NOTCH2 Recombinant Antibody (TAB-H67). The Goat Anti-Human IgG-HRP as a secondary antibody. Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.
