The anti-NOTCH2 pH-dependent antibody (VS-0825-YC268) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human NOTCH2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NOTCH2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NOTCH2 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human NOTCH2 is a transmembrane receptor of the Notch signaling pathway that plays a crucial role in immune cell development, particularly B-cells. Its dysregulation is linked to various diseases, including marginal zone lymphoma.
