The anti-CCR4 pH-dependent antibody (clone 1G1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CCR4. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CCR4 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CCR4 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CCR4 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CCR4 is a chemokine receptor expressed on the surface of various immune cells, particularly Th2 cells and regulatory T-cells. It is the receptor for CCL17 and CCL22, and its signaling is critical for the migration of these cells to sites of inflammation.
