SmartAb™ Recombinant Anti-CD3 pH-dependent Antibody (VS-0825-YC50)

This antibody binds specifically to human CD3 at acidic pH, but not at neutral or physiological pH, thereby selectively targeting the solid-tumor microenvironment. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD3 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD3 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD3 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD3 is a complex of T-cell receptor-associated proteins that are essential for T-cell signaling. It is comprised of CD3ε, CD3γ, CD3δ, and CD3ζ chains, which transduce signals from the T-cell receptor into the cell, triggering T-cell activation.
