Provided is an anit-CD3 single-domain antibody. The anit-CD3 single-domain antibody antagonizes the human CD3 epsilon N-terminal part, and the anit-CD3 single-domain antibody can be used for measurement materials such as CD3 and other functional protein requiring the CD3 such as a bispecific antibody.

SDS-PAGE analysis of HPAB-M0378-YC in β-mercaptoethanol-reduced (Lane 1) and non-reduced (Lane 2) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result of different β-mercaptoethanol-reduced and non-reduced conditions, the purified proteins migrate as about 11-17 kDa that is consistent with theoretical size.

Dot blot analysis of HPAB-M0378-YC was performed by loading human CD3 epsilon protein (Mouse IgG2a Fc Tag, 38.6 kDa).
HPAB-M0378-YC incubation concentration: 2 ng/μL.

Western blot analysis of HPAB-M0378-YC was performed by loading human CD3 epsilon protein (Mouse IgG2a Fc Tag, 38.6 kDa) onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for one hour. Membranes were probed with HPAB-M0378-YC (2 ng/μL) and HRP mouse anti-His tag as a secondary antibody (1: 2000) in turn. Chemiluminescent detection was performed.
Lane 1: Reducing antigen (0.1 μg)
Lane 2: Reducing antigen (0.3 μg)
Lane 3: Reducing antigen (0.6 μg)

ELISA analysis of HPAB-M0378-YC was performed by coating with human CD3 epsilon protein (Mouse IgG2a Fc Tag, 38.6 kDa). Then blocked with BSA and incubated with HPAB-M0378-YC. The HRP-conjugated mouse anti-His tag as a secondary antibody (1: 2000). Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.

Flow cytometry analysis of Jurkat cells incubated with anti-human CD3 recombinant antibody cat# HPAB-M0378-YC. Jurkat cells were stained with HPAB-M0378-YC (blue histogram, TE232382) or isotype control antibody (yellow histogram), followed by APC-conjugated goat anti-His tag secondary antibody. The used concentration of HPAB-M0378-YC was 10 µg per 5 x 10^6 cells.
