The anti-CD47 pH-dependent antibody (VS-0825-YC61) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD47. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD47 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD47 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD47 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD47 is a "don't eat me" signal protein ubiquitously expressed on the surface of most healthy cells. It binds to the SIRP alpha receptor on phagocytic cells, such as macrophages, to prevent them from being engulfed and destroyed. However, its overexpression in cancer cells allows them to evade destruction by the innate immune system.
