SmartAb™ Recombinant Anti-CD52 pH-dependent Antibody (Clone ANT1034)

The anti-CD52 pH-dependent antibody (clone ANT1034) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD52. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD52 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD52 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD52 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD52 is a small, glycosylphosphatidylinositol (GPI)-anchored protein found on the surface of lymphocytes and monocytes.
