The anti-CSF2 pH-dependent antibody (VS-0825-YC85) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CSF2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CSF2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CSF2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CSF2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CSF2, or Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF), is a cytokine that stimulates the production of granulocytes and monocytes. It plays a central role in hematopoiesis and is used clinically to stimulate blood cell production.
