The anti-CSF2RA pH-dependent antibody (VS-0825-YC371) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human colony stimulating factor 2 receptor, alpha, low-affinity (granulocyte-macrophage). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CSF2RA under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CSF2RA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CSF2RA is a subunit of the GM-CSF receptor that is crucial for hematopoiesis. Its signaling is essential for the proliferation and differentiation of myeloid cells, making it a potential therapeutic target.
