The anti-IL12A pH-dependent antibody (VS-0825-YC171) targets human IL12A in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL12A acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL12A clearance. The antibody operates in a cyclical manner, binding IL12A in the extracellular fluid and releasing it in acidified endosomes. Human IL12A is a subunit of Interleukin-12, a cytokine that is a key driver of the Th1 immune response. It is essential for the activation of T-cells and NK cells and is involved in the immune response against intracellular pathogens and cancer.
