The anti-IL15 pH-dependent antibody (VS-0825-YC174) targets human IL15 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL15 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL15 clearance. The antibody operates in a cyclical manner, binding IL15 in the extracellular fluid and releasing it in acidified endosomes. Human IL15, or Interleukin-15, is a cytokine that is crucial for the development, proliferation, and survival of NK cells and memory T-cells. It is a key player in The anti-tumor and anti-viral immune responses.
