The anti-IL4 pH-dependent antibody (VS-0825-YC202) targets human IL4 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL4 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. These histidine substitutions within its Complementarity-Determining Regions (CDRs) enable pH-responsive antigen binding, which leads to enhanced IL4 clearance. The antibody operates in a cyclical manner, binding IL4 in the extracellular fluid and releasing it in acidified endosomes. IL-4 (Interleukin-4) is a cytokine that is a central mediator of allergic responses. It plays a key role in the differentiation of T helper cells into Th2 cells, which promote B cell proliferation and the production of IgE antibodies.
